Enzymatic recycling assays provide a complementary approach for total glutathione. In these methods, glutathione reductase reduces oxidized glutathione while a thiol-reactive reagent, such as 5,5'-dithiobis(2-nitrobenzoic acid), produces a colored product. The reaction cycles between reduced and oxidized forms, amplifying the signal. Spectrophotometric or fluorometric detection can then estimate concentration. Distinguishing reduced glutathione from glutathione disulfide often requires separate aliquots, masking agents, or chromatographic separation, and the choice affects reported values.
Storage conditions strongly influence glutathione stability. The solid reduced form is commonly kept desiccated at or below minus twenty degrees Celsius, protected from light and moisture. Aqueous solutions are less stable because the thiol group reacts with dissolved oxygen, and oxidation accelerates at neutral or alkaline pH. Acidic solutions and oxygen-free handling can slow degradation, but repeated freeze-thaw cycles should be avoided. Researchers often verify concentration before use, because apparent losses can arise from oxidation or water uptake.
Quality control for glutathione measurements includes calibration with authenticated standards, internal standards where available, blank correction, and spike recovery checks. Because glutathione can form during sample processing or degrade before analysis, pre-analytical handling is a major source of variability. Interlaboratory comparisons often show differences in reported values due to method-specific calibration and detection principles. Interpretive thresholds are context-dependent, and no single reference range applies across all tissues or matrices. Researchers generally report both reduced and oxidized forms, along with the method and sample handling details.
Quantification of glutathione in biological or food samples commonly uses liquid chromatography coupled to ultraviolet, fluorescence, electrochemical, or mass spectrometric detection. Because the thiol group oxidizes readily, samples are often acidified or derivatized immediately after collection to stabilize reduced glutathione. Enzymatic recycling assays and colorimetric kits offer higher throughput but generally lower specificity than chromatographic methods. Mass spectrometry can distinguish glutathione from related thiols and allow simultaneous measurement of oxidized forms. Reported concentrations depend strongly on sample type, extraction procedure, and analytical platform.

Glutathione serves as a cofactor for several enzymes, including glutathione peroxidase and glutathione S-transferase. These enzymes help reduce hydrogen peroxide and lipid peroxides, and they conjugate reactive electrophiles for excretion. The molecule also acts as a reservoir for cysteine, an amino acid that is prone to oxidation. In addition, glutathione participates in the metabolism of nitric oxide, leukotrienes, and prostaglandins. Its roles extend to cell signaling, apoptosis, and the regulation of protein function through S-glutathionylation.
Glutathione is a tripeptide composed of glutamate, cysteine, and glycine, and it is the most abundant non-protein thiol in most living cells. The reduced form, GSH, carries a sulfhydryl group that can donate electrons, while the oxidized form, GSSG, forms when two GSH molecules link via a disulfide bond. The balance between these two forms helps define the cellular redox environment, and their ratio is often used as an indicator of oxidative stress. Because the sulfhydryl group is reactive, glutathione participates in many cellular processes, including detoxification and protein regulation.
Storage recommendations for glutathione reagents usually specify a cool, dry, dark environment because the thiol oxidizes in air and light. Solid material is often kept desiccated at low temperature, while solutions are prepared fresh or stored frozen in aliquots. Repeated freeze-thaw cycles can accelerate degradation, and metal ions can catalyze oxidation. Quality control may include purity assays, water content, and identity confirmation. Stability limits are method-specific, so a stated shelf life applies only to defined conditions and packaging.
Laboratory measurement of glutathione requires attention to oxidation before analysis. Blood, tissue, or cell samples can lose reduced glutathione as it converts to GSSG or forms mixed disulfides with proteins. Acid extraction, rapid freezing, and thiol-blocking reagents are common strategies to preserve the original distribution. Reported concentrations therefore depend on collection protocol, extraction method, and the time between sampling and analysis. Comparisons across studies are most reliable when these pre-analytical variables are described.
Common analytical approaches include enzymatic recycling assays, high-performance liquid chromatography, and mass spectrometry. The enzymatic recycling assay uses glutathione reductase and a colorimetric or fluorometric reagent to amplify signal, which gives good sensitivity for total glutathione. Chromatographic methods can separate GSH from GSSG and related thiols, while mass spectrometry offers structural confirmation and multiplexing. Each approach has different requirements for calibration, internal standards, and validation. No single method captures every form of glutathione in every matrix.
Samples for glutathione analysis require careful handling because the compound oxidizes readily and can be consumed by enzymes after collection. Blood is often treated with acid or thiol-blocking agents soon after draw, and plasma should be separated quickly from red blood cells. Tissues are usually snap-frozen or extracted immediately. Aqueous solutions of glutathione are less stable than dry powder and degrade faster at neutral or alkaline pH, in light, or with dissolved oxygen. Repeated freeze-thaw cycles also reduce reliability.
Quality control for glutathione materials checks identity, assay, purity, water content, and disulfide content. Commercial products vary from research-grade powder to dietary supplements, and labels may not distinguish reduced from oxidized forms. In the United States, oral glutathione is commonly sold as a dietary supplement rather than an approved drug, while injectable forms fall under different rules and may require a prescription. Regulatory status differs by country. Analytical certificates, when available, help verify what a material contains, but independent testing remains important for interpretation.
Laboratory measurement of glutathione typically starts with rapid acid extraction to prevent oxidation and enzymatic degradation. Common methods include enzymatic recycling assays, high-performance liquid chromatography, and liquid chromatography coupled with mass spectrometry. The recycling assay uses glutathione reductase and a thiol-reactive colorimetric or fluorescent reagent, measuring total glutathione after converting disulfide forms. Chromatographic methods can separate reduced and oxidized forms, which helps when the redox ratio is the target. Choice of method affects sensitivity, specificity, and the amount of sample needed.
== Structure == Pyruvic acid crystallizes as the keto acid, not the enol. The six non-hydrogen atoms are nearly coplanar. More relevant to biochemistry is the structure of the pyruvate anion. Several salts of pyruvate have been examined by X-ray crystallography. These tests confirm that pyruvate anion also exists in the keto form.
The lack of a separate chromosome 17 probe on the same section is an issue with regards to acceptance of CISH. As of June 2011 Roche has obtained FDA approval for the INFORM HER2 Dual ISH DNA Probe cocktail developed by Ventana Medical Systems. The DDISH (Dual-chromagen/Dual-hapten In-situ hybridization) cocktail uses both HER2 and Chromosome 17 hybridization probes for chromagenic visualization on the same tissue section. The detection can be achieved by using a combination of ultraView SISH(silver in-situ hybridization) and ultraView Red ISH for deposition of distinct chromgenic precipitates at the site of DNP or DIG labeled probes.

== History == Basilea Pharmaceutica was founded in 2000 as a corporate spin-off of the pharmaceutical company Roche. In 2002, Chinese subsidiary Basilea Pharmaceutica China Ltd. (BPC) was founded and was located in the Haimen Economic and Technological Development Zone in the city of Nantong in the Chinese province of Jiangsu, north of Shanghai. With the listing on the Swiss stock exchange in March 2004 at a price of CHF 98 per share, Basilea realised gross proceeds of more than CHF 200 million. In 2012, Basilea sold the worldwide rights to Toctino, a dermatology drug developed by Basilea that was approved and marketed in various European countries in 2008, to Stiefel Labs, a subsidiary of the British pharmaceutical company GlaxoSmithKline, for CHF 216 million. At the beginning of 2013, Basilea received orphan drug status in the USA for the antifungal drug Isavuconazole for the treatment of invasive fungal infections, which at the time, analysts estimated to be worth up to CHF 150 million per year. Isavuconazole was developed in phase III in conjunction with pharmaceutical company Astellas Pharma. In March 2015, Isavuconazole was approved in the US, followed by the entire EU in October of the same year. In the following years, the drug was also approved in Japan, several Eurasian countries, Australia and China.
A serial killer is typically a person who kills three or more people, with the murders taking place over more than a month and including a significant period of time between them. The Federal Bureau of Investigation (FBI) defines serial murder as "a series of two or more murders, committed as separate events, usually, but not always, by one offender acting alone". The United States has by far the largest number of documented serial killers in the world. According to Radford University's Serial Killer Information Center, it has more documented serial killers than the next ten highest countries on the list combined.

== Decaffeinated coffee == Friedlieb Ferdinand Runge performed the first isolation of caffeine from coffee beans in 1820, after the German poet Goethe heard about his work on belladonna extract, and requested he perform an analysis on coffee beans. Though Runge was able to isolate the compound, he did not learn much about the chemistry of caffeine itself, nor did he seek to use the process commercially to produce decaffeinated coffee.
=== Complications === Anorexia nervosa can have serious implications if its duration and severity are significant and if onset occurs before the completion of growth, pubertal maturation, or the attainment of peak bone mass. Complications specific to adolescents and children with anorexia nervosa can include growth retardation, as height gain may slow and can stop completely with severe weight loss or chronic malnutrition. In such cases, provided that growth potential is preserved, height increase can resume and reach full potential after normal intake is resumed. Height potential is normally preserved if the duration and severity of illness are not significant or if the illness is accompanied by delayed bone age (especially prior to a bone age of approximately 15 years), as hypogonadism may partially counteract the effects of undernutrition on height by allowing for a longer duration of growth compared to controls. Appropriate early treatment can preserve height potential, and may even help to increase it in some post-anorexic subjects, due to factors such as long-term reduced estrogen-producing adipose tissue levels compared to premorbid levels. In some cases, especially where onset is before puberty, complications such as stunted growth and pubertal delay are usually reversible. Anorexia nervosa causes alterations in the female reproductive system; significant weight loss, as well as psychological stress and intense exercise, typically results in a cessation of menstruation in women who are past puberty.
== Early life and education == Berhanu Kebede was born in Addis Ababa. He graduated in 1978 with a degree in economics from Addis Ababa University and continued his studies at the Free University of Brussels, earning M.A. degrees in Development Economics (1986) and in Management and Finance (1988).
== Biodistribution and mechanisms of action == Lithium compartmentalization was studied in 11 biosubstrates of rats when lithium ascorbate was taken at a dose of 1000 mg/kg. As part of a tubeless analysis of the dynamics of concentrations in whole blood, the following values of the pharmacokinetic parameters of lithium ascorbate were obtained: Cmax=50.59 µg/l, tmax=1.50 h, Clast=33.7 µg/l, AUCt=1750 µg/l*h, MRTt=22.9 h, Lz=0.005 1/h, T1/2=141 h, CL=0.029 l/h, Vd=5.9 l. Multichamber pharmacokinetic analysis showed that the stabilization of lithium levels in the blood is maintained by a special "depot" of lithium, apparently consisting of the aorta, femur and brain. Application of the method of functional linkage analysis suggested that the targeted accumulation of ascorbate anion in cells is due to the activity of vitamin C transport proteins SLC23A1, SLC23A2, SLC23A3. At the same time, lithium ions and ascorbate exhibit a synergistic effect. The main effects of lithium include the support of normal excitability central nervous system: by preventing excessive concentration of norepinephrine, regulation of sodium concentration in muscle cells (which is important for the tone of blood vessels), sensitivity of neurons to dopamine, which reduces the negative impact of stress.
It has become widely accepted in science that early in the history of life on Earth, prior to the evolution of DNA and possibly of protein-based enzymes as well, an "RNA world" existed in which RNA served as both living organisms' storage method for genetic information—a role fulfilled today by DNA, except in the case of RNA viruses—and potentially performed catalytic functions in cells—a function performed today by protein enzymes, with the notable and important exception of the ribosome, which is a ribozyme.

Evidence for the important role antithrombin plays in regulating normal blood coagulation is demonstrated by the correlation between inherited or acquired antithrombin deficiencies and an increased risk of any affected individual developing thrombotic disease. Antithrombin deficiency generally comes to light when a patient suffers recurrent venous thrombosis and pulmonary embolism.
=== Minimum requirement specification for Western Blot === In order to ensure that the results of Western blots are reproducible, it is important to report the various parameters mentioned above, including specimen preparation, the concentration of protein used for loading, the percentage of gel and running condition, various transfer methods, attempting to block conditions, the concentration of antibodies, and identification and quantitative determination methods. Many of the articles that have been published don't cover all of these variables. Hence, it is crucial to describe different experimental circumstances or parameters in order to increase the repeatability and precision of WB. To increase WB repeatability, a minimum reporting criteria is thus required.
=== Cochlear implants === In parallel to the development of neuromodulation systems to address motor impairment, cochlear implants were the first neuromodulation system to reach a broad commercial stage to address a functional deficit; they provide sound perception in users who are hearing-impaired due to missing or damaged sensory cells (cilia) in the inner ear. The approach to electrical stimulation used in cochlear implants was soon modified by one manufacturer, Boston Scientific Corporation, for design of electrical leads to be used in spinal cord stimulation treatment of chronic pain conditions.
In 2025, Medtronic was included in Forbes lists of America's Best Large Employers. In March 2026, Medtronic separated its diabetes business into MiniMed Group, Inc., which completed an initial public offering on March 9. Medtronic retained approximately 89.86% of MiniMed following the offering, while MiniMed became a separately publicly traded company on the Nasdaq. In September 2026, Medtronic launched an exchange offer as part of its planned separation of MiniMed. The offer covers up to 225,361,295 newly issued MiniMed common shares in exchange for Medtronic ordinary shares, representing approximately 80.1% of MiniMed's outstanding shares. If the exchange offer is oversubscribed, Medtronic intends to exchange additional shares of MiniMed representing its remaining interest, which would result in MiniMed becoming independent of Medtronic. The exchange offer is scheduled to expire on October 9, 2026, unless extended or terminated.

Chromatographic methods can separate the two forms before detection. Enzymatic assays often measure total glutathione first and then use a separate procedure to estimate the oxidized fraction. The difference between total and oxidized amounts provides an indirect estimate of the reduced form.
Acidification lowers pH and slows thiol oxidation during handling. It also helps precipitate proteins that could interfere with detection. Typical choices include metaphosphoric acid and sulfosalicylic acid.
Dissolved oxygen reacts with the thiol group, forming glutathione disulfide. Neutral and alkaline conditions generally increase the oxidation rate. Light, metal ions, and repeated freezing and thawing can also reduce stability.
