Several analytical methods can quantify glutathione, including high-performance liquid chromatography (HPLC) with UV or fluorescence detection for separating GSH and GSSG. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) offers higher specificity and sensitivity, often detecting nanomolar concentrations. The enzymatic recycling assay, often called the Tietze method, measures total glutathione by coupling reduction of GSSG to a colorimetric or fluorometric readout. Capillary electrophoresis and electrochemical detection are also used in specialized laboratories. Each method has distinct advantages and limitations regarding throughput, cost, and susceptibility to interference.
Interpreting glutathione measurements requires attention to pre-analytical variables. The GSSG concentration in a sample can rise artificially during storage or processing, making the GSH/GSSG ratio unreliable if not controlled. Reference ranges vary by specimen type, assay, and population, so comparisons across studies are difficult. Plasma glutathione is low and sensitive to hemolysis, while whole blood reflects primarily erythrocyte content. Many studies measure total glutathione rather than the reduced and oxidized forms separately, which limits conclusions about redox status.
Glutathione serves as a cofactor for several enzymes, including glutathione peroxidase and glutathione S-transferase. These enzymes help reduce hydrogen peroxide and lipid peroxides, and they conjugate reactive electrophiles for excretion. The molecule also acts as a reservoir for cysteine, an amino acid that is prone to oxidation. In addition, glutathione participates in the metabolism of nitric oxide, leukotrienes, and prostaglandins. Its roles extend to cell signaling, apoptosis, and the regulation of protein function through S-glutathionylation.
Glutathione is a tripeptide composed of glutamate, cysteine, and glycine, and it is the most abundant non-protein thiol in most living cells. The reduced form, GSH, carries a sulfhydryl group that can donate electrons, while the oxidized form, GSSG, forms when two GSH molecules link via a disulfide bond. The balance between these two forms helps define the cellular redox environment, and their ratio is often used as an indicator of oxidative stress. Because the sulfhydryl group is reactive, glutathione participates in many cellular processes, including detoxification and protein regulation.
Glutathione is synthesized in two ATP-dependent steps. First, gamma-glutamylcysteine synthetase links glutamate and cysteine; second, glutathione synthetase adds glycine to form the complete tripeptide. The pathway is feedback-inhibited by GSH itself, which helps maintain steady intracellular levels. Tissues vary widely in glutathione content, with the liver typically containing the highest concentrations, followed by the kidneys, lungs, and erythrocytes. Because cysteine is often limiting, its availability influences synthesis rates, and regulation of this pathway varies by cell type.

Cells synthesize glutathione through two ATP-dependent steps: glutamate-cysteine ligase joins glutamate and cysteine, and glutathione synthetase adds glycine to form the complete tripeptide. Breakdown occurs through gamma-glutamyl transpeptidase and subsequent peptidase reactions, forming the gamma-glutamyl cycle. Within cells, glutathione also forms a disulfide-linked dimer called GSSG when two GSH molecules react. The balance between GSH and GSSG is widely used as an indicator of oxidative conditions, although the ratio can vary by compartment and tissue.
Glutathione supports several cellular processes beyond direct antioxidant action. It serves as a cofactor for glutathione peroxidase and glutathione S-transferase enzymes, which reduce peroxides and conjugate electrophiles, respectively. The molecule also acts as a reservoir of cysteine, an amino acid that can limit protein synthesis and redox signaling. In human nutrition, oral glutathione is sold as a supplement, but how much intact glutathione reaches tissues after ingestion remains an active research question. Clinical claims about supplementation are not uniformly supported by controlled trials.
Stability depends on pH, temperature, oxygen exposure, and trace metals. Aqueous solutions of reduced glutathione are susceptible to oxidation, especially when neutral or alkaline and exposed to air. Transition metal ions can catalyze thiol oxidation, so chelators and inert atmospheres are sometimes used in research settings. Standards are typically stored cold and desiccated, with limited freeze-thaw cycles. Questions remain about how closely in vitro stability data reflect the behavior of glutathione within intact cells and tissues.
Measuring glutathione requires attention to oxidation during sample handling, because GSH in biological samples can convert to GSSG or form mixed disulfides with proteins after collection. Acidic extraction, rapid cooling, and chelating agents are commonly used to limit such changes. Analytical methods usually distinguish free reduced glutathione, total glutathione, and protein-bound forms. Because these forms have different stability and reactivity, reported values depend heavily on the preparation protocol. No single preparation is universally suitable for every biological matrix or analytical goal.
Some stellate cells were present in the macula flava, but started to show some signs of degeneration. The stellate cells synthesized fewer extracellular matrix molecules, and the cytoplasmic processes were shown to be short and shrinking, suggesting a decreased activity. Those results confirm the hypothesis that phonation stimulates stellate cells into producing more extracellular matrix. Furthermore, using a specially designed bioreactor, Titze et al. showed that fibroblasts exposed to mechanical stimulation have differing levels of extracellular matrix production from fibroblasts that are not exposed to mechanical stimulation. The gene expression levels of extracellular matrix constituents such as fibronectin, MMP1, decorin, fibromodulin, hyaluronic acid synthase 2, and CD44 were altered. All those genes are involved in extracellular matrix remodeling, thus suggesting that mechanical forces applied to the tissue, alter the expression levels of extracellular matrix related genes, which in turn allow the cells present in the tissue to regulate the extracellular matrix constituent synthesis, thus affecting the tissue's composition, structure, and biomechanical properties. In the end, cell-surface receptors close the loop by giving feedback on the surrounding extracellular matrix to the cells, affecting also their gene expression level.
== History == Elagolix was first described in the literature in 2005. It was originally developed by the pharmaceutical company Neurocrine Biosciences, and was later developed together by Neurocrine Biosciences and AbbVie (previously Abbott Laboratories). In June 2010, Neurocrine Biosciences and Abbott announced a global agreement to develop and commercialize elagolix for the treatment of endometriosis. The medication completed phase III clinical trials for endometriosis in November 2016. Two phase III clinical trials, the Elaris Endometriosis I and II (EM-I and EM-II) studies, were conducted. The studies included almost 1,700 women, about 950 of whom were treated with elagolix. In September 2017, AbbVie filed a New Drug Application (NDA) for elagolix for the treatment of pain associated with endometriosis in the United States. The medication was approved by the FDA for the treatment of endometriosis-associated pain in the United States on 23 July 2018. It was the first new medication to be approved by the FDA for the treatment of endometriosis in more than a decade. Elagolix was the first member of a new class of GnRH modulators described as "second-generation" due to their non-peptide and small-molecule nature and oral activity to be marketed. A second member of this group, relugolix (brand name Relumina), was introduced in Japan in January 2019. In addition to endometriosis, elagolix is under development for the treatment of uterine fibroids and menorrhagia. It is in phase III clinical trials for these indications.
== Description == The ESG may be performed with slight variations by different physicians. It most commonly is performed using the Apollo ESG Device (formally Overstitch device). This device fits over a therapeutic double-channel endoscope to create a sutured row of stomach tissue. The tissue helix is advanced from the endoscope, put up against stomach tissue, and turned 2-4 rotations to access the gastric muscle layer. The helix is then retracted towards the scope, bringing the full-thickness acquired stomach tissue with it. Using the handle-operated needle driver, a needle attached to the suture wire is passed through the full-thickness tissue to the anchor exchange. This creates a full-thickness plication of stomach tissue. The needle is then passed back from the anchor exchange to the needle driver, and the process is repeated, threading the suture wire through each bite of stomach tissue. When enough full-thickness bites have been taken for a suture row, a cinch is passed through the scope over the suture. Typically, sutures are placed starting at the border of the antrum and gastric body at the incisura, then placed proximally up to the border of the gastric body and fundus. Each row of sutures can be a straight line or one of the variety of suture patterns reported in the literature, such as the "M," "Z," and "U" pattern as well as other novel patterns; nonetheless, to date, no suture pattern has been proven to be superior for weight loss. Regardless of suture pattern, creation of the ESG focuses on tissue imbrication along the greater curvature of the stomach.
2009: Chinese geneticist Zeng Fanyi and her research team published their experiment results proving that induced pluripotent stem cells can be used to generate whole mammalian bodies – in this case, live mice.
The murder of James J. Smith occurred on November 20, 1932, when he was stabbed and bludgeoned to death by Allah Temple of Islam member Robert Karriem in what he described as a human sacrifice. Contemporary press labeled it the "Voodoo Murder". The killing led to the technical dissolution of the organization, which was reconstituted under the name Nation of Islam.

Specific vitamin A supplementation is particularly important for preventing further damage to the liver and skin. Ready-to-use therapeutic foods (RUTFs) and F-75 and F-100 milks were created to provide appropriate nutrition and caloric intake to those experiencing malnutrition. F-75 milk would be ideal when trying to reintroduce food into a malnourished person, and F-100 milk would be used to aid in weight gain. While RUTFs and F-100 milk were made to have the same nutritional value, RUTFs are beneficial as they are dehydrated and do not require much preparation. It is also important to note that infections are common in children with severe malnutrition and can further complicate treatment. Routine antibiotics, even in the absence of clinical infection, are generally given as a prophylactic measure, especially in regions with a high risk of infectious diseases. However, due to concerns about antibiotic resistance, there is debate over their routine use.
== History == Amino acid activation was first characterized by Mahlon Hoagland, who found that amino acids could be activated by certain enzymes to form an aminoacyl adenylate intermediate compound. The enzymes were discovered to be aminoacyl t-RNA synthetases, responsible for catalyzing the linkage of an amino acid to a small RNA molecule. Hoagland and his collaborator, Paul Zamecnik, later discovered that the small RNA molecule was a tRNA, and they identified it as a key facilitator of translation. Amino acid activation is a key reaction in many biochemical and metabolic processes. In particular, leucine natural killer treatment of interleukin-18 activated cells triggers the mTORC1 metabolic sensor, which indicates that mTORC1 causes amino acid-driven activation due to a high expression in amino acid transporters. The Inhibition of CD98/LAT1 amino acid transporters abroad mTORC1 activation driven by leucine, decreased the activity of the NK cell-effector.
=== Malema advertisement === During the South African national elections of 2009, Nando's made an advert lampooning African National Congress Youth League then president Julius Malema by using a puppet that resembled him. Malema's lawyers sued Nando's and the original advert was removed. However, an altered version was released, with the puppet's face pixelated and the voice altered. The puppet used in the advert was later sold at auction for 100,000 rand, which was donated to an educational charity.
Direct measurements of the Venusian atmosphere by spatial probes point to very harsh conditions, likely making Venus an uninhabitable world, even for the most extreme forms of life known on Earth. The extremely low water activity of the desiccated Venusian atmosphere represents the very limiting factor for life, much more severe than the infernal conditions of temperature and pressure, or the presence of sulfuric acid. Astrobiologists presently consider that more favorable conditions could be encountered in the clouds of Jupiter where a sufficient water activity could prevail in the atmosphere provided that other conditions necessary for life are also met in the same environment (sufficient supply of nutrients and energy in a non-toxic medium).

=== Post-translational proteolytic processing === Limited proteolysis of a polypeptide during or after translation in protein synthesis often occurs for many proteins. This may involve removal of the N-terminal methionine, signal peptide, and/or the conversion of an inactive or non-functional protein to an active one. The precursor to the final functional form of protein is termed proprotein, and these proproteins may be first synthesized as preproprotein. For example, albumin is first synthesized as preproalbumin and contains an uncleaved signal peptide. This forms the proalbumin after the signal peptide is cleaved, and a further processing to remove the N-terminal 6-residue propeptide yields the mature form of the protein.
=== Electrolyte replacement === Potassium replacement is often required as the metabolic problems are corrected. It is generally replaced at a rate of 10 mEq per hour as long as there is adequate urinary output.
The first aircraft carrying troopers from the 2–325th touched down at Point Salines 17 hours after H-Hour notification. In March 1988, a brigade task force made up of two battalions from the 504th Infantry and 3d Battalion (Airborne), 505th Infantry, conducted a parachute insertion and air/land operation into Honduras as part of Operation Golden Pheasant. The deployment was billed as a joint training exercise, but the paratroopers were ready to fight. The deployment caused the Sandinistas to withdraw to Nicaragua. Operation Golden Pheasant prepared the paratroopers for future combat in an increasingly unstable world.
=== Charges against UCC and UCIL employees === UCC chairman and CEO Warren Anderson was arrested and released on bail by the Madhya Pradesh Police in Bhopal on 7 December 1984. Anderson was taken to UCC's house after which he was released six hours later on $2,100 bail and flown out on a government plane. These actions were allegedly taken under the direction of then chief secretary of the state, who was possibly instructed from the chief minister's office, who himself flew out of Bhopal immediately. Later in 1987, the Indian government summoned Anderson, eight other executives and two company affiliates to appear in Indian court on homicide charges. In response, Union Carbide said the company is not under Indian jurisdiction. In May 1988, Indian businessman J. R. D. Tata wrote a confidential letter to the then Prime Minister, Rajiv Gandhi, conveying Henry Kissinger's concern about delay in reaching a settlement. At the time, Kissinger was an adviser to Union Carbide and other US corporations. Rajiv Gandhi indicated that he would consider Kissinger's advice. The opposition Bharatiya Janata Party alleged that Rajiv Gandhi protected Warren Anderson in a secret political deal with U.S. President Ronald Reagan, with Gandhi securing Reagan's pardon of Adil Shahryar in exchange. Dow was a named respondent in cases arising from Union Carbide's business in Bhopal.
In late 1999, al-Qaeda associate Walid bin Attash ("Khallad") contacted al-Mihdhar and told him to meet in Kuala Lumpur, Malaysia; al-Hazmi and Abu Bara al Yemeni would also be in attendance. The NSA intercepted a telephone call mentioning the meeting, al-Mihdhar, and the name "Nawaf" (al-Hazmi); while the agency feared "Something nefarious might be afoot", it took no further action. The CIA had already been alerted by Saudi intelligence about al-Mihdhar and al-Hazmi being al-Qaeda members. A CIA team broke into al-Mihdhar's Dubai hotel room and discovered that Mihdhar had a U.S. visa. While Alec Station alerted intelligence agencies worldwide, it did not share this information with the FBI. The Malaysian Special Branch observed the January 5, 2000, meeting of the two al-Qaeda members and informed the CIA that al-Mihdhar, al-Hazmi, and Khallad were flying to Bangkok, but the CIA never notified other agencies of this, nor did it ask the State Department to put al-Mihdhar on its watchlist. An FBI liaison asked permission to inform the FBI of the meeting but was told: "This is not a matter for the FBI."

Glutathione oxidizes quickly when cells are disrupted or when samples sit at room temperature. Rapid processing or immediate freezing minimizes the conversion of GSH to GSSG. This step helps ensure that the measured ratio reflects the original biological state.
The Tietze assay is an enzymatic recycling method that measures total glutathione. It uses glutathione reductase to reduce GSSG back to GSH, which then reacts with a chromogen or fluorophore. The reaction cycles repeatedly, amplifying the signal for detection.
Yes, but the choice of blood fraction matters. Plasma or serum contains low glutathione levels and is easily affected by hemolysis. Whole blood mainly reflects the high glutathione content of erythrocytes, so results from different fractions are not directly comparable.
Glutathione is a tripeptide of three amino acids: glutamate, cysteine, and glycine. The cysteine residue provides the sulfhydryl group that gives the molecule its reducing properties.
