Quality control for glutathione measurements includes calibration with authenticated standards, internal standards where available, blank correction, and spike recovery checks. Because glutathione can form during sample processing or degrade before analysis, pre-analytical handling is a major source of variability. Interlaboratory comparisons often show differences in reported values due to method-specific calibration and detection principles. Interpretive thresholds are context-dependent, and no single reference range applies across all tissues or matrices. Researchers generally report both reduced and oxidized forms, along with the method and sample handling details.
Quantification of glutathione in biological or food samples commonly uses liquid chromatography coupled to ultraviolet, fluorescence, electrochemical, or mass spectrometric detection. Because the thiol group oxidizes readily, samples are often acidified or derivatized immediately after collection to stabilize reduced glutathione. Enzymatic recycling assays and colorimetric kits offer higher throughput but generally lower specificity than chromatographic methods. Mass spectrometry can distinguish glutathione from related thiols and allow simultaneous measurement of oxidized forms. Reported concentrations depend strongly on sample type, extraction procedure, and analytical platform.
Synthesis occurs in two ATP-dependent steps. The enzyme glutamate-cysteine ligase joins glutamate and cysteine to form gamma-glutamylcysteine, and glutathione synthetase adds glycine. The first step is rate-limiting and is influenced by cysteine availability and feedback inhibition by GSH. Breakdown involves gamma-glutamyl transferase and subsequent peptidases, which release constituent amino acids for reuse. Because turnover differs among tissues, measurements from blood, plasma, and tissues are not directly interchangeable. Research continues to clarify how compartment-specific pools are regulated in health and disease.
Glutathione is a tripeptide composed of glutamate, cysteine, and glycine. It contains an unusual gamma-glutamyl bond between glutamate and cysteine, which resists cleavage by many peptidases. The reduced form, GSH, carries a thiol group on cysteine and is the dominant intracellular form in most cells. Its structure allows it to participate in redox reactions and to serve as a sulfur donor. The oxidized form, GSSG, consists of two GSH molecules joined by a disulfide bond.

Quality control for glutathione focuses on identity, purity, and oxidation state. Certificates of analysis may report assay value, water content, and the presence of GSSG or other impurities. Chromatographic purity is often expressed as a percentage of peak area. Reference standards help laboratories compare results across instruments and batches. Because glutathione is a small, polar molecule, separation from cysteine, gamma-glutamylcysteine, and related thiols can be challenging. Verification often combines more than one analytical technique.
Measuring glutathione requires attention to sample preparation because the molecule oxidizes readily. Blood, tissue, and cell samples are often treated with acid to precipitate proteins and stabilize the thiol. Without such steps, GSH can convert to GSSG or form mixed disulfides during storage. Analytical methods include spectrophotometric assays, high-performance liquid chromatography, and mass spectrometry. Each approach has different sensitivity, specificity, and susceptibility to interference from related compounds in complex matrices.
For solid glutathione, storage conditions affect shelf life. The reduced form is typically kept cool, dry, and protected from air and light. Moisture can promote oxidation, while elevated temperatures accelerate degradation. Suppliers often specify storage at or below freezing, sometimes under inert gas. Solutions are less stable than powders and may require preparation shortly before use. Buffers and chelating agents can slow oxidation, but they do not eliminate it. Published stability data vary with matrix, pH, and container.
Glutathione participates in detoxification reactions, amino acid transport, and the maintenance of protein thiols. It serves as a cofactor for several enzymes, including glutathione peroxidases and glutathione S-transferases. In research literature, altered glutathione status appears in studies of aging, infection, metabolic stress, and environmental exposure. Whether low glutathione is a cause, consequence, or marker of such conditions often remains unresolved. Direct measurement in blood or tissue provides a snapshot, but results depend on sample handling, timing, and the method used.
Glutathione is a small tripeptide made of glutamic acid, cysteine, and glycine. Its cysteine thiol group allows reversible oxidation and reduction, making it central to cellular redox chemistry. The reduced form, often abbreviated GSH, predominates inside most cells, while the oxidized disulfide form, GSSG, forms when two GSH molecules react. The ratio of GSH to GSSG is widely used as an indicator of oxidative stress in laboratory research, though it does not by itself diagnose a clinical condition.
Biosynthesis occurs in two ATP-dependent steps. The enzyme glutamate-cysteine ligase joins glutamate and cysteine, forming gamma-glutamylcysteine; glutathione synthetase then adds glycine to produce the complete tripeptide. Because the peptide bond from glutamate uses the gamma-carboxyl group, glutathione resists digestion by many ordinary peptidases. Tissues vary in synthesis capacity, and the liver generally contains high concentrations relative to many other organs. This uneven distribution contributes to organ-specific differences in redox buffering and affects how experimental results are interpreted across tissue types.
Storage conditions strongly influence glutathione stability. The solid reduced form is commonly kept desiccated at or below minus twenty degrees Celsius, protected from light and moisture. Aqueous solutions are less stable because the thiol group reacts with dissolved oxygen, and oxidation accelerates at neutral or alkaline pH. Acidic solutions and oxygen-free handling can slow degradation, but repeated freeze-thaw cycles should be avoided. Researchers often verify concentration before use, because apparent losses can arise from oxidation or water uptake.
Measuring glutathione in biological samples requires attention to oxidation and matrix effects. High-performance liquid chromatography with ultraviolet or fluorescence detection can separate reduced and oxidized forms after derivatization. Liquid chromatography with tandem mass spectrometry offers higher specificity and can quantify glutathione alongside related thiols. Because glutathione can oxidize during sample handling, many protocols use rapid acidification with metaphosphoric acid or sulfosalicylic acid. Internal standards help correct for losses during extraction and analysis.
Enzymatic recycling assays provide a complementary approach for total glutathione. In these methods, glutathione reductase reduces oxidized glutathione while a thiol-reactive reagent, such as 5,5'-dithiobis(2-nitrobenzoic acid), produces a colored product. The reaction cycles between reduced and oxidized forms, amplifying the signal. Spectrophotometric or fluorometric detection can then estimate concentration. Distinguishing reduced glutathione from glutathione disulfide often requires separate aliquots, masking agents, or chromatographic separation, and the choice affects reported values.
Interpreting glutathione measurements requires attention to pre-analytical variables. The GSSG concentration in a sample can rise artificially during storage or processing, making the GSH/GSSG ratio unreliable if not controlled. Reference ranges vary by specimen type, assay, and population, so comparisons across studies are difficult. Plasma glutathione is low and sensitive to hemolysis, while whole blood reflects primarily erythrocyte content. Many studies measure total glutathione rather than the reduced and oxidized forms separately, which limits conclusions about redox status.
Accurate measurement of glutathione begins with careful sample handling. Because GSH oxidizes rapidly to GSSG, samples must be processed quickly or frozen immediately. Acid precipitation with metaphosphoric acid or perchloric acid is common; it lowers pH, precipitates proteins, and helps preserve the reduced form. Chelating agents such as EDTA can limit metal-catalyzed oxidation. For whole blood, hemolysis releases glutathione from erythrocytes, so plasma and serum values differ substantially from whole blood values.
Several analytical methods can quantify glutathione, including high-performance liquid chromatography (HPLC) with UV or fluorescence detection for separating GSH and GSSG. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) offers higher specificity and sensitivity, often detecting nanomolar concentrations. The enzymatic recycling assay, often called the Tietze method, measures total glutathione by coupling reduction of GSSG to a colorimetric or fluorometric readout. Capillary electrophoresis and electrochemical detection are also used in specialized laboratories. Each method has distinct advantages and limitations regarding throughput, cost, and susceptibility to interference.
Afterwards, Adler takes extended time away from work, eventually revealing to Eric that he has a malignant brain tumor, and may not live longer than a year. As Pierpoint faces a debt crisis stemming from its ESG pivot, Adler clashes with senior leadership, particularly CFO Wilhelmina Fassbinder, over a potential sale of the firm, and brokers a meeting with Mitsubishi executives. Eric, realizing he needs to break free from Adler's influence, exploits Adler's illness during the meeting by gaslighting him into believing he overlooked a major error in the deal sheet. Adler has a breakdown and reveals his prognosis to the room; Eric escorts him out, and Adler realizes Eric betrayed him. Six months later, Adler has died, with Pierpoint organizing his memorial.
=== Interchangeability of centipede species in clinical practices === Comparative studies on S. mutilans and S. mojiangica, a common substitute for the former, has found that in pharmacodynamics and toxicology, the two centipede species have similar effects, and is theoretically proved that S. mutilans can be replaced by S. mojiangica. The same study found that two species showed similar results in various fields, including effects on animal haemoglobin and weight, chromosomal aberration tests, anticonvulsive tests, effects on in vitro fungal and bacterial growth, and acute toxicity tests. The study also suggested both centipede species can be used in relatively large doses, as they were both low in acute and genetic toxicities. Although the two centipede species were shown to be interchangeable in clinical practices, their interchangeability in clinical application was again questioned by some researchers, as more recent studies suggested that centipede proteins have a major role in their clinical effects. Through proteomics, researchers had found that the venoms of S. monjiangica and S. mutilans are different in the number of overlapping sequences and precursors for proteins and peptides. In addition, unique precursors were also found in both species for proteins and peptide levels. As the species differ in protein and peptide level, researchers of this study concluded that the interchangeability of centipede species requires further evaluation.

=== Heart failure === Cilostazol is dangerous for people with severe heart failure. Cilostazol has been studied in people without heart failure, without evidence of harm, but much more data would be needed to determine no risk exists. Although cilostazol would not be approvable for a trivial condition the Cardio-Renal Advisory Committee and FDA concluded that fully informed patients and physicians should be able to choose to use it to treat intermittent claudication. Patient and physician labeling will describe the basis for concern and the incomplete information available.

Mast cell degranulating (MCD) peptide is a cationic 22-amino acid residue peptide, which is a component of the venom of the bumblebee (Megabombus pennsylvanicus). At low concentrations, MCD peptide can stimulate mast cell degranulation. At higher concentrations, it has anti-inflammatory properties. In addition, it is a potent blocker of voltage-sensitive potassium channels.
== Food spoilage == Yeasts are able to grow in foods with a low pH (5.0 or lower) and in the presence of sugars, organic acids, and other easily metabolized carbon sources. During their growth, yeasts metabolize some food components and produce metabolic end products. This causes the physical, chemical, and sensible properties of a food to change, and the food is spoiled. The growth of yeast within food products is often seen on their surfaces, as in cheeses or meats, or by the fermentation of sugars in beverages, such as juices, and semiliquid products, such as syrups and jams. The yeast of the genus Zygosaccharomyces have had a long history as spoilage yeasts within the food industry. This is mainly because these species can grow in the presence of high sucrose, ethanol, acetic acid, sorbic acid, benzoic acid, and sulfur dioxide concentrations, representing some of the commonly used food preservation methods. Methylene blue is used to test for the presence of live yeast cells. In oenology, the major spoilage yeast is Brettanomyces bruxellensis. Candida blankii has been detected in Iberian ham and meat.
This was a boom year with a record high in exports, but the Scotch Whisky Association expressed concern for the future, particularly "the challenges posed by Brexit and by tensions in the global trading system". Scotch whisky tourism has developed around the industry, with distilleries being the third most visited attraction in Scotland; roughly 2 million visits were recorded in 2018. Some 68 distilleries operate visitors' centres in Scotland and another eight accept visits by appointment. Hotels, restaurants, and other facilities are also impacted by the tourism phenomenon. Tourism has had an especially visible impact on the economy in some remote rural areas, according to Fiona Hyslop MSP, Cabinet Secretary for Culture, Tourism and External Affairs: "The Scottish Government is committed to working with partners like the Scotch Whisky Association to increase our tourism offer and encourage more people to visit our distilleries." During the COVID-19 pandemic, exports of many food and drink products from the UK declined significantly, and that included Scotch whisky. Distillers were required to close for some time and the hospitality industry worldwide experienced a major slump. According to news reports in February 2021, the Scotch whisky sector had experienced £1.1 billion in lost sales. Exports to the US were also affected by the 25% tariff that had been imposed. Scotch whisky exports to the US during 2020 reportedly fell by 32% from the previous year. Worldwide exports fell in 70% of Scotch whisky's global markets.
=== Mechanism of action === Fialuridine (FIAU) is thymidine-like nucleoside analogue that was investigated as an antiviral against hepatitis B virus (HBV). HBV replication depends on its viral polymerase, which acts as reverse transcriptase and makes viral DNA. Because FIAU-TP resembles the normal nucleotide TTP, the viral polymerase can use FIAU-TP as an alternative substrate during DNA synthesis. This disrupts replication in two related ways. Fist, FIAU-TP can directly inhibit hepatitis virus DNA polymerase activity (shown in the duck hepatitis B virus model). Second, FIAU-TP interferes with the early protein-priming stage of reverse transcription. In an in-vitro priming system, FIAU-TP caused premature termination of the short starter DNA, which prevents proper extension into full-length viral DNA. Importantly, FIAU is not a classic obligate chain terminator, because it still has an intact 3’-OH group. However, once FIAU is incorporated into the growing DNA strand, extension becomes strongly slowed or blocked after a short step, so it behaves as a ‘de facto’ chain terminator. So FIAU's antiviral action can be summarized as follows: after FIAU is converted inside the cell into the active triphosphate form FIAU-TP, it can be used by the viral polymerase during BHV reverse transcription. This interaction disrupts the priming step and subsequent DNA chain synthesis, which eventually reduces the formation of complete viral DNA genomes.
In 1974, GM offered driver and passenger airbags as optional equipment on large Cadillacs, Buicks, and Oldsmobiles. In 1976, the crash test dummy Hybrid III was introduced to assess the impacts of car collisions. It represented the 50th percentile male standing at approximately 5' 9" tall and weighing 78 kg (171 lbs). In 1979, NHTSA began crash-testing popular cars and publishing the results, to inform consumers and encourage manufacturers to improve the safety of their vehicles. Initially, the US NCAP (New Car Assessment Program) crash tests examined compliance with the occupant-protection provisions of FMVSS 208. Over the subsequent years, this NHTSA program was gradually expanded in scope.

Bexagliflozin, sold under the brand name Brenzavvy, is an antidiabetic medication used to improve glycemic control in adults with type 2 diabetes. It is a sodium-glucose cotransporter 2 (SGLT2) inhibitor that is taken by mouth. The most common side effects include genital yeast infections, urinary tract infections, and increased urination. Bexagliflozin was approved for medical use in the United States in January 2023.
The UNESCO International Bioethics Committee recommended a precautionary global moratorium on the creation of mirror-image organisms, and the UK Government Office for Science held an expert roundtable that recommended "prevent[ing] the development of replicating mirror organisms." Several philanthropic funders have also stated that they will not fund research with the goal of creating mirror-image organisms, including Renaissance Philanthropy. In 2026, the UN Scientific Advisory Board called for "proactive multilateral action, such as a dedicated global forum, to define clear 'red lines', strengthen safety and monitoring practices, and establish responsible policy well before mirror life becomes feasible", and Chinese and US policy research organizations issued a joint statement calling for "national and international frameworks to guard against the risks posed by mirror life, reaffirming the shared view that such organisms should never be created." Some scientists and scholars have argued that concerns about mirror-image life are theoretical and/or that bans on research and funding bans are premature. Others have argued that the immune system might be able to recognize mirror versions of a certain type of biomolecule. Some scientists have proposed developing guidelines for synthetic biological entities regardless of their chirality.

=== Enzyme cyclization === Cyclization of enzymes by fusing the N- and C-termini of the protein helps elevate the stability of the enzyme against heat. By having SpyTag and SpyCatcher together in the enzyme at the termini, the enzyme will undergo spontaneous cyclization by forming the isopeptide bond. Cyclized beta-lactamase, phytase, firefly luciferase, and xylanase have shown retained enzyme activity following cooling, even after being subjected to heat at 100 °C.

Acidification lowers pH and helps prevent oxidation of the thiol group during extraction and storage. It can also precipitate proteins and stabilize the reduced form before analysis.
Blood contains glutathione, but concentrations differ between plasma and red blood cells. Careful separation and rapid processing are needed because ex vivo oxidation and hemolysis can alter results.
An enzymatic recycling assay uses glutathione reductase and a thiol-reactive reagent to generate a signal proportional to total glutathione. It is convenient for many samples but may not distinguish reduced and oxidized forms without additional steps.
GSH is the reduced form with a free thiol group, while GSSG is the oxidized disulfide-linked dimer. Most assays distinguish the two because their balance reflects redox conditions. The names are not interchangeable.
