=== Influence of preparation methods === Depending on the preparation and cooking methods, the nutritional value of potatoes can vary greatly. In particular, its energy content, moderate in comparison with other starchy foods, can increase considerably when cooked with fat, and its vitamin content is affected to a greater or lesser extent depending on the cooking method. However, cooking is essential to make it an appetizing and especially digestible food. In the raw potato, the starch is mainly in the form of resistant starch, so called because it resists digestive enzymes such as amylase. Under the effect of heat, around 50 °C, the amylose swells and causes the starch grains to burst, which "gelatinize" and lose their "resistant" character. However, when the preparation is subsequently cooled, e.g. in salads, the proportion of resistant starch increases due to a retrogradation of the amylose. In boiled potatoes, this proportion can be about 2% (of the total starch) and in potato salad it can be as high as 6%. The resistant starch remains intact in the large intestine, playing a role similar to that of dietary fiber, which may be of interest in some diets. A 100 g portion of potatoes simply boiled in their skins provides 76 kcal, which is comparable to corn porridge, also 76 kcal, or plantain (94), but is significantly lower than the same portion of dried beans (115), pasta (132), rice (135) or bread (278). ==== Soulja Boy ==== In June 2008, on DJ Cisco's Urban Legend mixtape, Ice-T criticized Soulja Boy (whose name is DeAndre Way) for "killing hip hop" and called his song "Crank That" "garbage" compared to the works of other hip-hop artists such as Rakim, Das EFX, Big Daddy Kane and Ice Cube. One of the comments exchanged was Ice-T telling Way to "eat a dick". The two then traded numerous videos back and forth over the Internet. Trump ==== A lawsuit was filed on February 6, 2025, by the American Foreign Service Association and the American Federation of Government Employees in the US District Court for the District of Columbia, requesting a temporary restraining order and preliminary injunction against the administration, claiming that it violated separation of powers, the Take Care Clause of the Constitution, and the Administrative Procedure Act and requesting that all attempts to shut down the agency be halted, all recent actions be reversed, and a new acting director be appointed. The following day, U.S. District Judge Carl Nichols, nominated by President Trump in 2019, stated from the bench that he would enter in a "temporary restraining order", pausing the plan to put thousands of employees on leave and pausing the accelerated removal of workers from their posts abroad. On February 21, 2025, Judge Nichols cleared the way for the Trump administration to move forward with pulling thousands of USAID staffers off the job in the United States and around the world, as part of an administration plan to also provide those abroad with a 30-day deadline to move back to the U.S. at government expense. Nichols had previously argued that Trump's actions threaten the safety of USAID workers abroad because many are deployed in unstable regions.
== Indications and Usage == In 1997, the United States Food and Drug Administration approved difloxacin hydrochloride tablets for use in canines. Unlike other fluoroquinolones difloxacin has not been approved by the FDA for use in cats. It is also contraindicated for use in food animals. The recommended dosing is 5–10 mg/kg once daily. This is based on the linear pharmacokinetics exhibited by difloxacin over the 5–60 mg/kg range. So the maximum dose is well within the established safety range, but drug accumulation in tissues prompts a recommendation that the medication should not be used for periods longer than thirty days. If there is no noticeable improvement after five days, then the condition should be reevaluated and all therapy options reconsidered. Difloxacin is indicated for infections caused by susceptible microorganisms in the conditions listed below.

Possibly the most common use of affinity chromatography is for the purification of recombinant proteins. Proteins with a known affinity are protein tagged in order to aid their purification. The protein may have been genetically modified so as to allow it to be selected for affinity binding; this is known as a fusion protein. Protein tags include hexahistidine (His), glutathione-S-transferase (GST), maltose binding protein (MBP), and the Colicin E7 variant CL7 tag. Histidine tags have an affinity for nickel, cobalt, zinc, copper and iron ions which have been immobilized by forming coordinate covalent bonds with a chelator incorporated in the stationary phase. For elution, an excess amount of a compound able to act as a metal ion ligand, such as imidazole, is used. GST has an affinity for glutathione which is commercially available immobilized as glutathione agarose. During elution, excess glutathione is used to displace the tagged protein. CL7 has an affinity and specificity for Immunity Protein 7 (Im7) which is commercially available immobilized as Im7 agarose resin. For elution, an active and site-specific protease is applied to the Im7 resin to release the tag-free protein.

=== 2024 Khabarovsk, Russia === On 5 April 2024, an emergency regime was introduced in the Russian city of Khabarovsk after a local resident accidentally discovered that radiation levels had jumped sharply in one of the industrial areas of the city. According to volunteers of the dosimetric control group, the dosimeter at the NP site showed up to 800 microsieverts, which is 1600 times the safe value. Employees of the Ministry of Emergency Situations fenced off the area to 30 by 30 meters (100 by 100 ft), where they found a capsule with caesium from a defectoscope. This was placed in a protective container and taken away for disposal. The incident was first reported by the Novaya Gazeta.
